Drie Musketiers volgt de avonturen van de jonge d'Artagnan die naar Parijs reist om zich aan te sluiten bij de Koninklijke Garde. Na een tumultueus begin raakt hij bevriend met de drie Musketiers, Athos, Porthos en Aramis, die samen leven onder het motto 'Eén voor allen, allen voor één'. De groep wordt al snel betrokken bij een web van intriges en gevaren aan het hof van de koning, waarbij ze hun vaardigheden met de sabel moeten gebruiken om hun eigen leven en de eer van Frankrijk te beschermen. De roman is een klassiek voorbeeld van de historische avonturenroman, vol actie, romantiek en avontuur.
