In der düsteren Atmosphäre des Dartmoor trifft auf Sherlock Holmes und Dr. Watson ein außergewöhnlicher Fall. Sir Charles Baskerville wird tot aufgefunden, sein Gesicht von Entsetzen gezeichnet und am Tatort finden sich Spuren eines riesigen Hundes. Der Legende nach soll der „Hund der Baskervilles“ auf dem Moor sein Unwesen treiben, seit ein Vorfahre Sir Hugos ein Mädchen zu Tode hetzte und daraufhin von dem dämonischen Hund getötet wurde. Als der Erbe des Anwesens eintrifft, erhält er einen anonymen Brief, der ihn eindringlich vor dem Moor warnt. Holmes und Watson müssen ihre detektivischen Fähigkeiten aufbieten, um das Geheimnis des Hundes zu lüften und die Beteiligten zu entlarven.
