Hester Prynn, een jonge vrouw die veroordeeld is tot het dragen van een scharlaken letter 'A' op haar kleding als teken van haar adulterie, wordt gedwongen haar leven in het puritaanse Boston te leiden onder de veroordelende blikken van haar medemensen. Haar minnaar, de predikant Arthur Dimmesdale, ondergaat een innerlijke strijd tussen schuld en begeerte, terwijl hij zijn ware identiteit verborgen houdt. De verhaallijn wordt complexer wanneer Hester's echtgenoot, Roger Chillingworth, verschijnt en wraak zoekt op de persoon die hem heeft verraden.
